Abstract : Background: The tetracyclic compounds of C-4-hydroxyphenylcalix [4] pyrogalolarene (Pg4OH) are synthetic compounds that show strong antioxidant properties that can be developed as topical antioxidants to neutralize free radicals due to UV exposure. One of the steps of drug development is toxicity testing. For topical preparations, one of the tests that must be carried out is an acute skin irritation test. Objective: This study was conducted to determine the skin irritation effect of Pg4OH. Research Methods: This quasi experimental with posttest only design carried out in vivo on 3 males albino adult rabbits. All 3 rabbits whose hair on the back was shaved. This shaving was done 24 hours before being treated. The Pg4OH was exposed for 4 hours then observed at 24, 48 and 72 hours after exposure to see the erythema and edema and was scored. The result of the observation was calculated with the primary irritation index which then the calculation result was categorized by Globally Harmonized System (GHS) and International Organization for Standardization (ISO) categories. Results: Among three rabbits only one rabbit showed erythema and edema with very small size (score: 1) at 72 hours at 3 sites ie 1 control site and 2 sample sites with each value was 1. Two other rabbits obtained 0 value at each observation location. The results of the calculation of the primary irritation index from the observation data obtained were 0,0093 included in the category of very light or negligible. Conclusion: The tetracyclic compound Pg4OH does not irritate the skin.
