Abstract : Background: Uric acid (UA) plays important role in inflammation in the joint. Recently, uric acid also induces inflammation and fibrosis in kidney, and allopurinol treatment reduces the diseases progression. However, its role in other organ, especially liver has not been completely investigated. This study elucidated effect of allopurinol administration after uric acid induction liver function and fibrosis in association with TGF- β1 and Hepatic Growth Factor (HGF) expression. Methods: Hyperuricemia was induced in mice using intraperitoneal injection of UA 125 mg/Kg body weight (BW) and NaCl injection in control group. Mice were sacrificed on days-7 (UA7) and 14 (UA14). Allopurinol was administered with dose 50mg/Kg BW orally for 7 days after uric acid induction (U7AL7 and U14AL7). Liver function was assessed from serum SGOT and albumin level. Liver was harvested and used for RNA extraction and paraffin making. Reverse Transcriptase-PCR (RT-PCR) was performed for quantifying TGF-β1, Collagen-1 and HGF. Paraffin section was used for Sirius red (SR) and immunohistochemical (IHC) staining. Results: Uric acid induction caused liver injury with higher SGOT and lower albumin levels. Meanwhile, allopurinol administration induced lower SGOT with higher albumin level compared to uric acid treated groups. Sirius red and immunohistochemical staining showed slight collagen and myofibroblast deposition in interstitial areas of UA treated group, which associated with higher TGF-β1 and Collagen-1 mRNA expression with downregulating HGF mRNA expression. In other hand, allopurinol administration revealed downregulation of TGF-β1 and Collagen-1 mRNA, with upregulation of HGF mRNA expression. Conclusions: Allopurinol administration after uric acid induction attenuate liver injury and fibrosis.
