Abstract | : | Green tea is one type of tea which extensively used to treat many kind of disease such as hypertension. Green tea have a high contain antioxidant called catechin.Catechin can act as a radical scavenger for free radicals such as singlet oxygen, hydroxyl radicals and peroxynitrite. Based on its benefits, catechincan eliminate heavy metal i.e lead in human body, especially for lead-exposed individual i.e car autorepair shop workers. Chelation is the treatment of choice in lead poisoning. This treatment usually use a chemical chelating agent likedimercaprol, but it has side effect like hypertension. The objective of these studies were to know (1) the effect of green tea on NO level in leadexposed individual with NOS3 gene polymorphism and non polymorphisms (2) The effect of green tea on systolic and diastolic in lead-exposed individual with polymorphisms gene NOS3 and non polymorphisms. The studies using a survey method with a cohort design. These subject were 30 car autorepairs hop workers as cases subject and 30 control subject from Pamijenvillage.The genotype individu NOS3 genepolymorphisms investigated by Polimerase Chain Reaction (PCR)- Restriction Fragment Length Polymorphisms (RFLP) with MBoI restricted enzyme. Parameter recorded were blood lead, nitric oxide level. systolic and diastolic blood pressure. Data were analyzed by pair t-test. These results showed that 40% of 30 cases subject were detected as polymorphisms individual of gene NOS3, with GA genotype. Mbo-I enzyme cut their DNA become 260 bp, 119 bp and 87 bp. As many as 60% of 30 cases subject were detected as non polymorphisms individual with GG genotype. NO level both on the individual of polymorphisms gene NOS3 and non polimorphisms were increased and their systolic and diastolic blood pressures were decreased. This research can be concluded that green tea has an activity as a chelator agent for lead |
