Abstract : 4-hydroxychalcone is a chalcone derivative that can be an antioxidant and can thus be used as an active ingredient in sunscreen UV-B absorption. This research aimed to determine the levels of carbopol, CMC-Na, and gelatin that could generate the optimum formula for 4-hydroxychalcone gel, to test the physical properties, and to determine the behavior of UV-B absorption of sunscreen in vitro and in vivo. We calculated the optimum formula on the physical properties of the gel using the Simple Lattice Design method with Design-Expert version 10.1 software. Data analysis with a one-sample t-test was performed to determine the variations in the results of the physical properties test of the optimum formula generated in the software and the results of the observations. Sunscreen activity was seen from the SPF value collected from in vitro UVVis spectrophotometer and in vivo tests using New Zealand White female rabbits. The findings revealed that the proportion of carbopol, CMC-Na, and gelatin that provided the optimum formula for 4-hydroxychalcone gel was 1%; 2.5 %; and 1.5 %. Physical properties such as dispersion forces, adhesion, and viscosity of the results observations in the software did not vary significantly. The in vitro activity of 4-hydroxychalcone sunscreen gave a Sun Protection Factor (SPF) value of 21.49 and in vivo resulted in an SPF value of 21. The optimum formula for 4-hydroxychalcone gel preparation had moderate UV-B absorption sunscreen activity
